Abstract

Based on DNA barcoding technology and COI sequences from the GenBank database,a systematic analysis was conducted on the species composition of amphioxus in the Huangcuo sea area of Xiamen.A total of 48 amphioxus specimens were examined,yielding COI se-quence fragments with a length of 492 bp.Through BLAST homology comparison in the GenBank database and phylogenetic tree construction,the results revealed 37 specimens of Branchiostoma japonicum(77%)and 11 specimens of B.belcheri(23%).These findings indicate that the amphioxus population in the Huangcuo sea area of Xiamen is a mixed group,consisting of both B.japonicum and B.belcheri,with B.japonicum being the current dominant species.This discovery updates the traditional understanding that the area was primarily inhabited by B.belcheri,while also suggesting potential ecological niche differentiation between the two species.Morphological analysis showed that the amphioxus popu-lation in Xiamen is predominantly composed of medium to large individuals,with such individuals accounting for 37.8%in the B.japonicum group and 27.3%in the B.belcheri group.AMOVA analysis indicated no significant differences in the mean body length and body weight be-tween the two species(P>0.05).The results of this study provide a scientific basis for the targeted conservation and management of amphioxus resources in the Xiamen sea area.
