摘要

Abstract

The microwave-assisted extraction conditions were optimized through orthogonal experiments.This study used the national certified reference material(clam quality control sample)as raw material.A three-factor three-level orthogonal experimental design L9(33)was employed to determine inorganic arsenic extraction conditions,combined with liquid chromatography-atomic fluorescence spectrometry(LC-AFS)for inorganic arsenic quantification.The orthogonal experiment results revealed that the optimal extraction conditions were:nitric acid volume 14mL,nitric acid concentration 0.12 mol/L,extraction temperature110℃,and extraction time 25 minutes.The theoretical inorganic arsenic content in clam was 0.62 mg/kg,while the measured content reached 0.60 mg/kg with a relative deviation of 2.4%.The standard recovery rate reached 92.0%~95.0%,demonstrating the accuracy and reliability of this orthogonal experimental model.Compared with traditional hot leaching methods,this approach offers advantages such as shorter processing time,lower solvent usage,and operational convenience and rapidity.This study provides scientific basis for clam detection research and quality safety control of aquatic products.
