摘要

Abstract

This study aims to establish a tissue culture rapid propagation technology system for the sugarcane Chuantang102183,to screen the optimal culture media and cultivation substrates throughout the callus culture process,and to provide technical support for the industrialization of healthy sugarcane seedlings.Stem tip heart leaves of Chuantang102183 were used as explants for rapid propagation via plant callus culture technology.The optimal media for callus induction,advantitious bud differentiation,and root growth stages,as well as the most suitable cultivation substrate for the sham planting stage,were screened.The optimal medium for callus induction of Chuantang102183 was MS+3.0 mg/L 2,4-D,and its induction rate was 86.11%.The most suitable advection differentiation medium was MS+2.5 mg/L 6-BA+0.2 mg/L NAA,and the most suitable rooting medium was 1/2MS+2.0 mg/L IBA,with a rooting rate of 100%.The most suitable substrate for false planting seedlings was a mixture of peat soil and perlite(4∶1),resulting in an average survival rate of 85.22%.This study successfully established a full-process tissue culture system for Chuantang102183 from callus induction,indefinite bud differentiation,root growth,transplantation.Based on callus culture technology,this system provides an innovative approach for the large-scale production of healthy sugarcane seedlings and has the potential for industrial promotion.
