Isolation and identification of culturable fungi from rice false smut balls in Nanchang region,Jiangxi Province

Abstract
[Objective]Rice false smut is a kind of fungal disease seriously harming rice worldwide.Rice false smut ball is a spherical structure,formed after the infection of Ustilaginoidea virens in rice panicle,with rich nutrients,and provides favorable conditions for the growth of a variety of microorganisms,especially suitable for fungal survival.Studying the diversity of culturable fungi in rice false smut balls can lay a foundation for exploring the interaction between Ustilaginoidea virens and other fungi as well as rice.[Method]The rice false smut ball was divided into the outer layer(dark green chlamydospore layer),middle layer(yellow mature mycelium layer)and inner layer(white tender mycelium layer).The culturable fungi were separated from these three parts by tissue separation method.The types of fungi were identified based on morphological characteristics and molecular biology.[Result]A total of 32 fungi strains were isolated from rice false smut balls by the tissue separation method.The 32 strains of fungi were identified as 15 genera by morphological observation and rDNA-ITS sequence analysis.Among them,12 strains of fungi were isolated from the outer layer of the rice false smut balls and identified as the following 8 genera:Ustilaginoidea,Bjerkandera,Apiospora,Gelatoporia,Penicillium,Fusarium,Aspergillus and Trametes,among which Aspergillus had three strains and was the dominant population.A total of 11 strains of fungi were isolated from the middle layer of rice false smut balls and identified as the following 8 genera:Ustilaginoidea,Alternaria,Curvularia,Cladosporium,Aspergillus,Epicoccum,Lecanicillium and Clonostachys,among which Alternaria had three strains and were the dominant population.A total of 9 strains of fungi were isolated from the inner layer of the rice false smut balls and identified as the following 6 genera:Ustilaginoidea,Fusarium,Cladosporium,Alternaria,Talaromyces,and one unidentified fungal.Three strains of Cladosporium were isolated,which meant Cladosporium was the dominant population.In general,Aspergillus,Alternaria and Cladosporium showed high isolation frequency.[Conclusion]The population and diversity of culturable fungi in various parts of rice false smut balls in Nanchang area of Jiangxi Province were preliminarily clarified,which laid a foundation for further researches on the interaction of different fungi during the formation of rice false smut balls and the abundant sources of secondary metabolites in rice false smut balls.关键词
