Characterization of the COBRA Gene Family in Catalpa bungei and Its Transcriptional Response in Tension Wood Formation

Abstract
[Objective]This study aimed to conduct a genome-wide identification and comprehensive char-acterization of the COBRA gene family members(CbuCOB)in Catalpa bungei,and to elucidate their tran-scriptional response patterns during tension wood(TW)formation.[Methods]Bioinformatic approaches were employed to identify and systematically analyze the CbuCOB genes.RNA sequencing was used to investigate the expression profiles of CbuCOB genes across different tissues and during TW formation in C.bungei.Quantitative real-time PCR(qRT-PCR)was further conducted to validate the expression levels of key CbuCOB genes.[Results]A total of 14 CbuCOB genes were identified in the C.bungei genome,exhibiting an uneven chromosomal distribution.Phylogenetic analysis classified these CbuCOB proteins into two subfamilies(SF I and SF II)and 12 subgroups,with members within the same subgroup exhibit-ing high structural similarity.Promoter cis-element analysis revealed that CbuCOB genes contain multiple elements associated with growth and development,environmental stress responses,and plant hormone signaling.Tissue-specific expression analysis indicated that CbuCOB genes exhibited distinct expression patterns in different tissues,with CbuCOB1,CbuCOB2,CbuCOB3,and CbuCOB9 showing higher expres-sion levels in xylem.Temporal transcriptome analysis showed dynamic transcriptional responses of Cbu-COB genes during TW formation,with five SF I members(CbuCOB1,CbuCOB2,CbuCOB8,CbuCOB9,and CbuCOB12)and one SF II member(CbuCOB4)identified as differentially expressed genes.qRT-PCR validation confirmed that the expression trends of key CbuCOB genes were consistent with the transcrip-tome results,suggesting that these genes may be involved in regulating TW formation in C.bungei.[Con-clusion]The COBRA gene family in C.bungei exhibits diverse gene characteristics and expression pat-terns,with several members likely playing key important roles in wood formation,particularly during TW formation.关键词
