A Mendelian randomization study of the causal association between COVID-19 and lung cancer risk

Abstract
Objective:To explore the causal relationship between coronavirus disease 2019(COVID-19)and the risk of developing lung cancer.Methods:A two-sample Mendelian randomization analysis was employed.Genetic instrumental variables for COVID-19 severity,hospitalization,and susceptibility phenotypes were obtained from large-scale genome-wide association studies.The causal effects of COVID-19 on overall lung cancer risk and on the risks of specific subtypes(lung adenocarcinoma,lung squamous cell carcinoma,and small cell lung cancer)were assessed using the inverse variance weighted(IVW)method as the primary analysis,supplemented by four other MR methods.The robustness of the results was verified through heterogeneity tests,pleiotropy tests,and MR-PRESSO analysis.Results:IVW analysis showed that neither the COVID-19 severity phenotype(OR=1.013,95%CI:0.953-1.077,P=0.679),hospitalization phenotype(OR=1.056,95%CI:0.961-1.159,P=0.257),nor susceptibility phenotype(OR=1.131,95%CI:0.856-1.494,P=0.388)increased the overall risk of lung cancer.The results from the other four MR methods(MR-Egger,weighted median,simple mode,weighted mode)were consistent with the IVW method.Analysis of lung cancer subtypes also indicated no causal association between any of the COVID-19 phenotypes and subtype risks(all P>0.012 5).Sensitivity analyses revealed no significant heterogeneity or pleiotropy.Conclusion:No genetic evidence was found to support an increased risk of lung cancer due to COVID-19.This conclusion requires further validation by prospective clinical studies.关键词
