基于指纹图谱和多指标含量测定结合化学计量学的不同产地北苍术药材质量评价研究
Quality evaluation of Rhizoma Atractylodis Chinensis from different origins based on finger-print profiles,multi-indicator content determination and chemometrics

Abstract
Objective To establish HPLC fingerprints and multi-component content determination methods for Rhizoma Atractylodis Chinensis and to evaluate the quality of Rhizoma Atractylodis Chinensis from different origins by chemical pattern recognition.Methods A C18 chromatographic column(4.6 mm×150 mm,5 µm)was used as the stationary phase,acetonitrile(A)and 0.2%phosphoric acid solution(B)were used as the mobile phase for gradient elution,and common peaks were confirmed by comparing with reference substances.Subsequently,fingerprint common peak analysis and similarity evaluation were conducted on 15 batches of Rhizoma Atrac-tylodis Chinensis medicinal materials,and the characteristic components of Rhizoma Atractylodis Chinensis were screened by cluster analysis(CA),principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA).Mean-while,the contents of atractylenolide II,atractylodinol,β-eucalyptol and atractylodin in 15 different sources of Rhizoma Atractylodis Chinensis were measured for comparative analysis.Results A total of 11 common peaks were identified in the fingerprint,with similarity greater than 0.820.Chemical pattern analysis showed that the compounds atractylenolide II,atractylodinol,β-eucalyptol,and atracty-lodin could be used as characteristic components for the quality evaluation of the medicinal material.Specifically,there were differences in the content of Rhizoma Atractylodis Chinensis among different origins,the content of atractylodin in S6 sample was the highest,the total amount of 4 chemical components in Sample S12 was the highest,and the differences in the total content of the four chemical com-ponents among S1-S5 samples were small and the content was relatively stable.Conclusion The established HPLC fingerprint method and multi-component content determination method are suitable for the quality evaluation of Rhizoma Atractylodis Chinensis from differ-ent sources,and the results of content determination,fingerprint analysis and chemical pattern analysis of different origins can provide a basis for the selection of origin and quality control of Rhizoma Atractylodis Chinensis medicinal materials.关键词
