沙棘叶多糖的分离、结构表征及生物活性研究
Study on the Separation,Structural Characterization and Biological Activity of Polysaccharides from Hippophae rhamnoides L.Leaves

Abstract
To provide a reference for the comprehensive utilization of sea buckthorn(Hippophae rhamnoides L.).In this study,seabuckthorn leaves were taken as the research object,and a neutral(HRLP-Ⅰ)and an acidic polysaccharide(HRLP-Ⅱ)were isolated through water extraction and alcohol precipitation method,mac-roporous resin,and DEAE Sepharose Fast Flow anion exchange column chromatograph,and their structural fea-tures and in vitro antioxidant activity were investigated.Further analysis on the immunomodulatory effects of HRLP-Ⅱ on the spleen of immunosuppressed mice was evaluated in vivo.The results showed that HRLP-Ⅰcontained more glucose and galactose,while the dominant monosaccharides of HRLP-Ⅱ were galactose and arabinose,both of which were heteropolysaccharides with typical polysaccharide infrared absorption peaks.The advanced structural characterization results showed that there was no triple helix structure for both HRLP-Ⅰ and HRLP-Ⅱ,and exhibited different surface morphology features.Thermochemical analysis indicated that HRLP-Ⅱhad better thermal stability than HRLP-Ⅰ.The in vitro antioxidant results showed that 2,2'-azino-bis(3-ethyl-benzothiazoline-6-sulfonic)(ABTS),1,1-diphenyl-2-picrylhydrazyl(DPPH),iron ion chelation and total reduc-ing of HRLP-I were superior to those of HRLP-Ⅱ,while the hydroxyl radical scavenging ability of HRLP-Ⅱwas superior to that of HRLP-Ⅰ.Compared with the model group,HRLP-Ⅱ high-dose group(400 mg/kg BW),medium-dose group(200 mg/kg BW)and low-dose group(100 mg/kg BW)could significantly reduce the expression of IL-2,IL-6 and TNF-α in the spleen of mice(P<0.05),decrease the lesions of node hyperpla-sia and red bone marrow congestion in the spleen tissue of mice and increase the ratio of CD4+/CD8+,reduce the lesions of T lymph in the spleen of mice.In summary,the HRLP-Ⅱ separated from sea buckthorn leaves can play an immunomodulatory role by regulating the cyine expression of spleen cells in immunosuppressed mice,im-proving the lesions of spleen tissue and T lymphocyte balance.关键词
