Abstract

The objective of this study is to systematically identify the zincfinger protein(ZFP)gene family in Verticillium dahliae,characterize its evolutionary features,and investigate the expression profile of VdZFP dur-ing host infection.Based on the whole genome sequence of V.dahliae,ZFP genes were identified using bioinfor-matics tools.Comprehensive analyses were performed,including phylogenetic tree construction,gene structure annotation,chromosomal distribution,and subcellular localization prediction.The expression pattern of VdZFP during cotton infection further validated via qRT-PCR.A total of 77 ZFPs genes were identified in V.dahliae.Phylogenetic analysis classified them into five distinct subfamilies(Ⅰ~Ⅴ),each showing significant structural and functional divergence.Chromosomal mapping revealed a non-random clustered distribution of these genes.Subcellular localization predictions indicated that 82%of ZFPs genes were nuclear-localized.qRT-PCR results demonstrated that the members of the VdZFP family may play an important role in the infection process.Among them,VdZFP16 exhibited a unique bimodal expression pattern-early suppression,followed by late activa-tion during infection,with expression down regulation to 33%of the control at 4-8 hpi and sharply up-regulated to10.57-fold at 48 hpi.These findings suggest that VdZFP16 may facilitate fungal pathogenesis through its bi-modal expression dynamics and could serve as a potential target for controlling Verticillium wilt.This study pro-vides important insights into the functional roles of ZFP genes in V.dahliae and their mechanisms during patho-gen-host interaction.
