Exploring the medication patterns and action mechanisms of national patent Chinese medicated diets in regulating diabetes based on data mining and network pharmacology

Abstract
Objective To investigate the medication patterns and action mechanisms of national patent medicated diets of Chinese medicine in regulating diabetes mellitus(DM)based on data mining and network pharmacology.Methods Authorized medicine-food homologous compound formulas were collected from the official website of the China National Intellectual Property Administration.Microsoft Excel 2016,SPSS Modeler 18.0,and R Studio were utilized to perform frequency statistics,property-flavor-meridian affinity analysis,association rule mining,and cluster analysis,respectively.The"herb-compound-target"network was constructed using Cytoscape 3.10.2 to predict core drug targets and action pathways.Molecular docking verification was subsequently conducted using AutoDock Tools 1.5.7 and PyMOL.Results(1)A total of 136 medicine-food homologous compound formulas were collected,involving 83 medicine-food homologous herbs.The total frequency of herb usage was 1,269.Twenty-eight high-frequency herbs(used≥10 times)were identified.Warm,neutral,and cold properties predominated,while sweet,pungent,and bitter flavors were most frequent.The herbs primarily acted on the spleen,stomach,lung,liver,and kidney meridians.Association rule mining and cluster analysis revealed the core medicinal combination to be Shanyao(Dioscoreae Rhizoma),Gegen(Puerariae Lobatae Radix),Gouqizi(Lycii Fructus),Huangjing(Polygonati Rhizoma),and Fuling(Poria).(2)Network pharmacology analysis indicated that the main active compounds are quercetin,β-sitosterol,diosgenin,3'-methoxydaidzein,and stigmasterol.The core targets were identified as AKT1,tumor necrosis factor(TNF)-α,interleukin(IL)-6,TP53,and IL-1β,involving signaling pathways such as AGE-RAGE and IL-17.Molecular docking verification confirmed favorable binding activity between the core compounds and targets(ΔG≤-5.0 kcal/mol).Conclusion Chinese medicated diets regulate DM by exerting effects such as strengthening the spleen,tonifying qi,and promoting fluid production to quench thirst.This is achieved through core herbs including Shanyao(Dioscoreae Rhizoma),Gegen(Puerariae Lobatae Radix),Gouqizi(Lycii Fructus),etc.,which act on targets like AKT1,TNF-α,and IL-6,and modulate signaling pathways such as AGE-RAGE and IL-17.关键词
