轻悦饮料对肥胖相关抑郁症的改善作用及其分子机制初探
Preliminary exploration of the ameliorative effects and molecular mechanisms of Qingyue Beverage on obesity-associated depression

Abstract
Objective To investigate the antidepressant effect of Qingyue Beverage[composed of Meiguihuɑ(Rosae Rugosae Flos),Jiangxiangru(Moslae Herba),Baishao(Paeoniae Radix Alba),Chenpi(Citri Reticulatae Pericarpium),and Bohe(Menthae Haplocalycis Herba)]on mice and to preliminarily elucidate its molecular mechanisms in ameliorating obesity-associated depression based on network pharmacology and molecular docking.Methods The antidepressant activity of Qingyue Beverage was evaluated using the tail suspension test,open field test,and sucrose preference test.Network pharmacology was employed to screen the active compounds and targets of the formula,and a"compound-target-pathway"network was constructed.A protein-protein interaction(PPI)network was built using the STRING database,and core targets were identified through Cytoscape 3.9.1.Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis was performed on the Metascape platform.Finally,molecular docking of core compounds with key targets was validated using AutoDock and PyMOL.Results Behavioral experiments showed that Qingyue Beverage significantly reduced the immobility time in the tail suspension test(P<0.05),and increased the central grid movement time while decreased the central grid residence time in the open field test(P<0.01).Network pharmacological screening identified 55 active compounds(e.g.,quercetin,acacetin,luteolin)and 316 potential targets.Core targets included AKT1,PPARG,catalase(CAT),interleukin(IL)-1β,and FOS,which were primarily enriched in pathways such as pathways in cancer,PI3K-Akt signaling pathway and HIF-1 signaling pathway.Molecular docking confirmed favorable binding activity between the core compounds and key targets(ΔG≤-5.0 kcal·mol-1).Conclusion Qingyue Beverage may exert antidepressant effects and alleviate obesity-associated depression through the synergistic actions of multiple compounds(e.g.,quercetin,luteolin)on multiple targets(e.g.,AKT1,PPARG),thereby modulating related signaling pathways such as PI3K-Akt.This mechanism demonstrates the characteristic"holistic regulation"of compound formulas of Chinese medicines.关键词
