Abstract
In order to identify the prevalence of avian astro virus(AAstV)in waterfowl in China,1 351 tissues and swab samples collected from ducks and geese in 15 provincial-level administrative regions from 2023 to 2024 were detected for pathogen nucleic acids using a quadruple fluorescent quantitative PCR assay,from which 16 AAstV-positive samples were randomly selected for ORF2 sequencing and genetic evolution analysis.The results revealed that 235 samples were positive for AAstV nucleic acids,with an overall positivity rate of 17.39%,including 47 positive for GoAstV-1,61 for GoAstV-2,89 for DAstV-3 and 75 for DAstV-4,of which,37 samples were identified as co-infection.Positive samples were detected from all the 15 regions,in particular,the eastern and southern coastal regions where detection rate was higher than that in the central and northern inland regions;the site-level positivity rate was high in wholesale markets and farms/households compared to that in farmer's markets,while no positive sample was detected in slaughterhouses;and the positivity rate in geese(31.15%)was significantly higher than that in ducks(11.04%).The types of prevalent AAstV were different depending on different species population,specifically,DAstV-4 and GoAstV-2 were dominant in geese,while DAstV-3 was in ducks;all the ORF2 genes of the 16 representative AAstV-positive samples clustered in the same clade as their respective reference sequences,and were highly identical to the reference strains.In conclusion,it was possible that AAstV infection was widely spread in waterfowl in China,particularly intensive farming areas with high risk of transmission.It was thus necessary to control any prevalence of AAstV through improving biosafety management on poultry farms.关键词
