贵阳市1株G9P[13]型猪轮状病毒的分离鉴定与遗传进化分析
Isolation,Identification and Genetic Evolutionary Analysis of One Strain of G9P[13]Type Porcine Rotavirus in Guiyang City of Guizhou Province

Abstract
In order to confirm the cause of diarrhea affecting a pig herd on a farm in Guiyang City,fecal samples collected from diarrheic pigs were detected for relevant viruses that were subsequently isolated and identified,followed by genetic evolutionary analysis of the virus genes.The results showed that the positivity rate was 67.5%(54/80)for porcine rotavirus(PoRV),but negative results were obtained for porcine epidemic diarrhea virus(PEDV),transmissible gastroenteritis virus(TGEV)and porcine deltacoronavirus(PDCoV).The positive samples were processed and inoculated into MA-104 cells,where stable cytopathic effects appeared after 36 hours.Specific fluorescence was observed by indirect immunofluorescence assay,and one strain of PoRV was successfully isolated.It was found that,by phylogenetic analysis of VP4 and VP7 genes of the isolated strain,the VP4 gene was 96.9%identical to P[13]type murine-derived strain RVA/Murine-tc/CHN/SCLS-M/2018/G3P[13](MK606442);and the VP7 gene was 96.6%homologous to G9 type porcine-derived strain China/GZ/2023(PQ323320),and 88.8%to the U.S.G9 type human-derived strain OM46(AJ491181).Ultimately,the isolated strain was identified as G9P[13]type,and genetically correlative to rotavirus of murine or human origin.A reference was provided for future epidemiological researches and prevention of PoRV infection in China.关键词
