2株GⅥ-1基因型鸡传染性支气管炎病毒的分离鉴定及致病性研究
Isolation,Identification and Pathogenicity Characterization of Two Strains of GⅥ-1 Genotype Infectious Bronchitis Viruses

Abstract
In order to identify the molecular characteristics and pathogenic properties of emerging strains of infectious bronchitis virus(IBV),clinical samples suspected of IBV infection submitted by Hunan and Guizhou Province were subjected to virus isolation and identification,and S1 gene phylogenetic analysis and pathogenicity testing in specific-pathogen-free(SPF)chickens were carried out.The results showed that two IBV strains were successfully isolated and designated as CK/CH/HN/K2305 and CK/CH/GZ/Q2444,respectively;regular death and stunted growth,including characteristic lesions such as dwarfed and curled embryos,could be induced by the two strains when they were inoculated into 9 to 11-day-old SPF chicken embryos.The full-length of the S1 genes were 1 624 bp and 1 617 bp,respectively;phylogenetic analysis revealed that both the strains were classified into GⅥ-1 genotype,the homology was 96.4%between their S1 gene nucleotides,ranged from 75.3%to 78.3%with GⅥ-1 reference strain,but from 53.6%to 54.4%with GⅠ-19(QX)genotype reference strain prevalent in China.Pathogenicity testing based on 2-day-old SPF chicks revealed that the mortality rates caused by CK/CH/HN/K2305 and CK/CH/GZ/Q2444 were 40%and 10%,respectively,by both of which,pathological damages at different levels could be induced in tracheas,lungs,kidneys and bursa of fabricius of the infected chicks;and viral shedding monitoring showed that the positive detection rates of both the oropharyngeal and cloacal swabs were 100%when the challenged and contact-exposed groups were during the experiment period for 15 days.The pathogenic characteristics and S1 gene genetic evolution of GⅥ-1 genotype IBV were analyzed in the study,enriching the biological database of the virus,and providing valuable data for future prevention and control against the disease.关键词
