摘要

Abstract

The aim of this study is to investigate the function of DNA double-strand break repair protein RAD51 in the replication of pseudorabies virus(PRV).Cell lines from various species were infected with PRV,and the RAD51 expression following PRV infection was analyzed by Western-blot,RT-PCR and RT-qPCR.RAD51 overexpression and knockdown A549 cells were generated by lentivirus packaging systems,and the replication of PRV in control and RAD51 overexpression or knockdown cell lines was examined.Meanwhile,the effects of RAD51 specific inhibitors(IBR,B02 and DIDS)on the replication of PRV were evaluated.Western-blot,RT-PCR and RT-qPCR results showed that PRV infection significantly downregulated the expression of RAD51 in various host cells.Western-blot,RT-PCR analysis showed that the PRV replication was significantly enhanced in RAD51 overexpression cell line compared with control cell line,while depletion of RAD51 significantly inhibited the viral replication.In addition,Western-blot,RT-qPCR and RT-PCR data showed that treatment with three RAD51 inhibitors(IBR2,B02 and DIDS)significantly reduced the PRV replication in cells.Collectively,these findings reveal a critical role of RAD51 in facilitating the replication of PRV,and provided new insights into the interaction between PRV infection and host DNA damage response.
