Abstract

This study aimed to investigate infectious bursal disease virus(IBDV)-induced cellular pyroptosis and its underlying mechanisms.Cell viability was assessed using the CCK-8 assay,cell death types were analyzed via flow cytometry,and cell membrane integrity was evaluated by lactate dehydrogenase(LDH)release.The results showed that cell viability significantly decreased(P<0.01),the proportion of Annexin-V/PI double-positive cells increased,and LDH release was elevated at 36 h post-infection.GSDME mRNA expression was significantly upregulated at 24 h post-infection(P<0.05),indicating the occurrence of pyroptosis.Although transfection with VP2 or VP5 alone did not significantly induce LDH release,both reduced cell viability(P<0.001).Following IBDV infection,VP2 or VP5 transfection further exacerbated cell damage,suggesting their potential to enhance pyroptosis.Mdivi-1 treatment mitigated pyroptosis,implying that IBDV may regulate this process via the mitochondrial pathway.These findings provide new insight into the pathogenic mechanisms of IBDV.
