Abstract

To develop a recombinant live vectored vaccine capable of effectively preventing porcine reproductive and respiratory syndrome(PRRS)and porcine parvovirus(PPV)infection,the reverse genetics platform of the PRRS virus(PRRSV)live vaccine was utilized.The gene encoding the protective antigen protein VP2 of PPV was inserted between the structural and non-structural proteins of PRRSV,constructing a full-length infectious clone of pPRRSV-PPV-VP2.The recombinant virus rPRRSV-PPV-VP2 was rescued in Marc-145 cells,and its plaque morphology,genetic stability,and viral replication kinetics were compared with those of the parental virus.Additionally,piglets were immunized with the recombinant virus to evaluate its ability to elicit immune responses in pigs.The results showed that the VP2 gene of PPV could be stably inherited and efficiently expressed in the recombinant virus rPRRSV-PPV-VP2.The clinical performance of piglets immunized by the recombinant virus rPRRSV-PPV-VP2 was normal,and the autopsy and pathological sections showed that there were no abnormal pathological changes in various tissues and organs,which had good safety for piglets,and at the same time,it could induce piglets to produce specific antibodies against PRRSV and PPV at the same time,which had good immunogenicity.The serum samples of all test pigs on the 0,7th,14th,21st,28th and 35th day after immunization showed that the levels of VP2 protein antibody and N protein antibody turned positive on 7 days after immunization,and the levels of the two antibodies reached their peaks on 21 and 28 days,respectively.The above results indicate that rPRRSV-PPV-VP2 is a prospective live viral vector vaccine against PRRSV and PPV.
