奶牛源黏液型多杀性巴氏杆菌的分离鉴定及其生物学特性分析
Isolation,identification and biological characterization of mucoid Pasteurella multocida from cows

Abstract
To determine the biological characteristics of mucinous Pasteurella multocida and its differences from that of nonmucinous P.multocida,three Gram-negative and Pasteurella multocida-like bacterial isolates,including one mucoid isolate YN241063 and two nonmucoid isolates YN241034 and YN241011 were isolated from lungs of cows with respiratory diseases in Yunnan Province.The three isolates were identified by biochemical analysis and 16S rDNA gene sequencing.The differences in biological characteristics,including capsular serotyping,multi-locus sequence typing(MLST),pathogenicity and antimicrobial susceptibility,of the three isolates were analyzed.The results of bacterial identification showed that the most biochemical characteristics of the three isolates were identical to those of the P.multocida reference strains HNZM01 and CVCC44801.The 16S rDNA gene sequence similarities between the three isolates and P.multocida type strain ATCC 43137T was from 99.6%to 99.9%.The three isolates formed the same clade with all the P.multocida reference strains in the phylogenetic tree of the 16S rDNA gene.Based on the above characteristics,all the three isolates were identified as P.multocida.The capsular serotypes of the three P.multocida isolates YN241063,YN241034 and YN241011 were A,A and D,respectively,and the MLST types were ST3,ST178 and ST133,respectively,and the median lethal doses(LD50)in mice were 2.5 × 103 CFU,4.0 × 103 CFU and 1.0 × 104 CFU,respectively.The mucoid isolate YN241063 was resistant to streptomycin,kanamycin,gentamicin,amikacin,and sulfamethoxazole,but the two nonmucoid isolates YN241034 and YN241011 were sensitive to all 14 antimicrobial agents.As far as we know,this is the first isolation of mucoid P.multocida in China,confirming the presence of mucoid P.multocida infection in cows in China,and which providing important reference data for the laboratory diagnosis,and prevention and control of the mucoid P.multocida infections in cows in China.关键词
