Fertility Recovery and Sperm Ultrastructure of Male Nile Tilapia with gsdf Gene Deletion after Letrozole Treatment

Abstract
[Objective]Deletion of the gonadal soma-derived factor(gsdf)in Nile tilapia(Oreochromis niloticus)results in reduced male fertility and sexual reversal to females.This study aims to analyse sperm ultrastructure and investigate whether fertility can gradually recover over time,providing foundational data for elucidating the regulatory mechanisms of gsdf in germ cell development and maturation during spermatogenesis in teleost fish.[Methods]The Nile tilapia XY gsdf-/-individuals were fed with the diet containing 200 μg/g aromatase inhibitor letrozole(LE)for 3 months,thereafter their sex were restored as male.Following sexual maturation,regular artificial insemination trials were conducted to continuously compare fertilization rates between LE-treated gsdf-deficient(XY gsdf-/-LE)and wild-type(XY gsdf+/+LE)males.Concurrently,scanning electron microscopy and transmission electron microscopy were used to compare sperm morphology and ultrastructure among XY gsdf-/-LE,XY gsdf+/+LE,and untreated wild-type control individuals(XY gsdf+/+Ctrl).[Results]At the age of 9 to 21 months,the fertilization rate of XY gsdf-/-LE individuals was found to be considerably lower in comparison to that of XY gsdf+/+LE individuals(P<0.05).Consequently,the fertilization rate exhibited a gradual increase and approached a level of XY gsdf+/+LE individuals at the age of 24 to 30months(P>0.05).Electron microscopy revealed that at 18 months of age,sperm morphology of XY gsdf-/-LE individuals resembled that of the control XY gsdf+/+group,both exhibiting the fundamental structural characteristics of teleost sperm,and no apparent structural abnormalities were observed.[Conclusion]The fertility of male tilapia lacking the gsdf gene,which is restored through LE rescue,can recover with age,and the early reduction in fertility is not caused by damage to sperm ultrastructure.关键词
