基于网络药理学和分子对接探讨乙酰紫堇灵通过类泛素化修饰在胃癌中的可能作用机制
The potential action mechanism of acetylcorynoline on gastric cancer via SUMOylation based on network pharmacology and molecular docking

Abstract
Objective:To explore the potential action mechanism of acetylcorynoline on gastric cancer via SUMOylation based on network pharmacology and molecular docking.Methods:Potential targets related to acetylcorynoline,SUMOylation,and gastric cancer were mined from multiple databases and online platforms.The intersection targets of the three were taken to construct a drug-SUMOylation-disease network diagram.The GO function and KEGG pathway enrichment analyses were performed by the Metascape platform.The GEPIA database was used to verify the differential expression of core targets in gastric cancer tissues and adjacent tissues,as well as their correlation with prognosis.The TIMER database was employed to validate the correlation of ALB and TGF-B1 with tumor immune infiltration cells.The binding of acetylcorynoline with key targets was simulated online by CB-Dock2 and visualized.Results:A total of 178 potential targets of acetylcorynoline,2 421 SUMOylation-related targets,and 2 029 gastric cancer-related targets were selected.According to the Venn tool,57 intersection targets were obtained.Ten core targets,namely CASP3,STAT3,ALB,ESR1,EGFR,MTOR,HSP90AA1,ANXA5,PTGS2,and TGF-B1,were identified through the protein-protein interaction network.The GO and KEGG enrichment analyses indicated that these targets were associated with signaling pathways such as cancer-related pathway,PI3K-Akt signaling pathway,cellular senescence,and cellular apoptosis.It was found that the expression levels of ALB and TGF-B1 in gastric cancer tissues and normal gastric tissues were different and associated with overall survival.TGF-B1 expression level was positively correlated with the level of immune cell infiltration.Molecular docking results showed that acetylcorynoline had good affinities and stable interactions with the core targets ALB and TGF-B1.Conclusion:Acetylcorynoline may exert anti-gastric cancer effects by regulating multiple links such as signaling pathways including cancer-related pathways,PI3K-Akt signaling pathway,cell senescence,cell apoptosis through regulating the SUMOylation of genes such as ALB and TGF-B1.关键词
