The action mechanism of integrated stress response on Alzheimer's disease based on the theory of"Spleen deficiency and phlegm turbidity"
摘要
Abstract
Alzheimer's disease is a neurodegenerative disorder characterized pathologically by the deposition of β-amyloid protein plaques and hyperphosphorylation of tau protein.Accumulating evidence indicates that neurons can continuously respond to dynamic cellular environments.Alzheimer's disease,as an aging-associated neurodegenerative disorder,shows a declined ability to adapt to the homeostatic mechanisms.Integrated stress response,a signal central control network triggered by protein homeostasis disorder primarily through modulating protein synthesis rates,plays a significant role in the pathogenesis of Alzheimer's disease.Soluble β-amyloid protein oligomers,the main hallmark of Alzheimer's disease,trigger integrated stress response through multiple pathological pathways including neuronal hyperexcitation,endoplasmic reticulum stress,oxidative stress,and amino acid deprivation.The theory of"spleen deficiency and phlegm turbidity"is one of the key pathogenic mechanisms of Alzheimer's disease in traditional Chinese medicine(TCM),wherein spleen deficiency serves as the initiating factor in Alzheimer's disease pathogenesis,and phlegm turbidity represents as a pivotal factor in the disease progression.The process of spleen deficiency leading to phlegm turbidity is highly consistent with the action mechanism of integrated stress response in the pathogenesis of Alzheimer's disease.In this paper,the aberrant activation and regulatory mechanisms of the integrated stress response in Alzheimer's disease are explored from the perspective of"spleen deficiency and phlegm turbidity",aiming to provide novel insights for the clinical management of Alzheimer's disease.关键词
