Abstract
Objective:Based on network pharmacology and molecular docking technology,the material basis and action mechanism of Fufang Siyadan Shengfa Ding(复方斯亚旦生发酊)in the treatment of androgenetic alopecia(AGA)were explored initially to provide a theoretical foundation for its clinical application.Methods:The ingredients of the three Chinese medicinal materials in Fufang Siyadan Shengfa Ding and their corresponding targets were collected by TCMSP,CNKI,PubMed,Swiss Target Prediction databases.GeneCards,DisGeNET,OMIM,and TTD databases were used to collect the targets related to AGA,so as to obtain the common targets of disease and medicines,and then GO and KEGG enrichment analyses were performed.The Cytoscape was used to construct the protein-protein interaction network and the"medicine-ingredient-target-pathway"network,and the main active ingredients of Fufang Siyadan Shengfa Ding and the core targets were verified by molecular docking.Results:A total of 316 common targets have been identified based on the intersection of active ingredients of Fufang Siyadan Shengfa Ding and potential disease targets,among which the key targets included HSP90AA1,STAT3,SRC,ESR1,JUN and so on.The biological processes in GO enrichment analysis mainly included peptidyl tyrosine modification,positive regulation of MAPK cascade,positive regulation of kinase activity,etc.KEGG enrichment analysis mainly included signaling pathways such as PI3K/Akt,MAPK,Ras-related protein 1,and chemokines.The molecular docking results showed that the core targets had good binding abilities with most of the active ingredients in Fufang Siyadan Shengfa Ding.Conclusion:The mechanism of Fufang Siyadan Shengfa Ding in the treatment of AGA may be to promote cell proliferation,regulate the cell cycle,and inhibit apoptosis by mediating the expression of relevant proteins in the PI3K/Akt and MAPK signaling pathways through a variety of ingredients such as kaempferol,apigenin,luteolin,sinapinic acid,and quercetin.关键词
