miR-140-5p调控IGF1R/mTOR信号通路对炎症微环境中人牙周膜干细胞成骨分化的影响
The effect of miR-140-5p on osteogenic differentiation of human periodontal ligament stem cells in inflammatory microenvironment by regulating the IGF1R/mTOR signaling pathway

Abstract
Objective To explore the effect of miR-140-5p on osteogenic differentiation of human periodontal ligament stem cell(HPLSC)in inflammatory microenvironment by regulating the insulin-like growth factor 1 receptor(IGFlR)/mammalian target of rapamycin(mTOR)signaling pathway.Methods HPLSC were treated with 5.0μg/ml lipopolysaccharide to induce inflammatory microenvironment and separated into model group,inhibitor NC group,inhibitor miR-140-5p group,inhibitor miR-140-5p+si-NC group,and inhibitor miR-140-5p+si-IGF1R group.HPLSC cultured normally were served as control group.Quantitative real-time polymerase chain reaction was performed to measure miR-140-5p and IGF1R mRNA.5-ethynyl-2'-deoxyuridine(EdU)was used to measure cell proliferation.Alkaline phosphatase staining was used to measure osteogenic differentiation.Alizarin red staining was performed to observe the formation of mineralized nodules in cells.Western blotting was performed to detect the expression of IGF1R/mTOR signaling pathway proteins.The dual luciferase reporter gene was used to analyze the targeting relationship between miR-140-5p and IGF1R.Results Compared with control group,model group showed an increase in miR-140-5p level,a decrease in IGF1R mRNA level,EdU positivity rate,alkaline phosphatase activity,reduced formation of mineralized nodules,and a decrease in IGF1R and p-mTOR protein levels(P<0.05).Compared with inhibitor NC group,inhibitor miR-140-5p group showed a decrease in miR-140-5p level,an increase in IGF1R mRNA level,EdU positivity rate,alkaline phosphatase activity,an increase in mineralized nodule formation,and an increase in IGF1R and p-mTOR protein levels(P<0.05).Compared with inhibitor miR-140-5p+si-NC group,inhibitor miR-140-5p+si-IGF1R group showed a decrease in IGF1R mRNA level,EdU positivity rate,alkaline phosphatase activity,mineralized nodule formation,and IGF1R and p-mTOR protein levels(P<0.05).The dual luciferase reporter gene experiment showed that miR-140-5p had a targeted relationship with IGF1R(P<0.05).Conclusion MiR-140-5p may inhibit osteogenic differentiation of HPLSC in the inflammatory microenvironment by suppressing the IGF1R/mTOR signaling pathway.关键词
