Abstract
Objective To construct a prognostic model related to trefoil factor 3(TFF3)for lung adenocarcinoma,and analyze the predictive value of this model for immunotherapy.Methods Using the TIMER,UALCAN and TISCH databases,the expression of TFF3 and its relationship with the stage of lung adenocarcinoma,as well as the cellular localization within lung adenocarcinoma tissues,were analyzed.Based on the HPA database,the expression of TFF3 in lung adenocarcinoma tissues was analyzed from the perspective of immunohistochemistry.Based on the expression level of TFF3,lung adenocarcinoma patients were divided into low expression group and high expression group.Then,a comparative analysis of TFF3-related differential genes was conducted.Univariate survival analysis and LASSO-Cox regression were used to construct the TFF3-related risk score.The receiver operating characteristic curve was employed to evaluate the predictive efficacy of this risk score,and a prognostic nomogram for lung adenocarcinoma was established.Results The expression of the TFF3 gene and protein in lung adenocarcinoma tissues were significantly higher than those in normal tissues(P<0.05).Moreover,the higher the stage of lung adenocarcinoma,the higher the expression of the TFF3 gene and protein.The single-cell data analysis of lung cancer showed that TFF3 was mainly expressed in tumor cells.The 12 genes identified through univariate survival analysis were subjected to LASSO-Cox regression screening,and an 8-gene TFF3-related prognostic gene signature was constructed.The corresponding risk score was calculated.The overall prognosis of patients with lung adenocarcinoma in low-risk group was better than that in high-risk group(P<0.001).The immune score of patients in high-risk group was lower than that in low-risk group,suggesting that patients in high-risk group had a poorer response to immunotherapy.The prognostic nomogram established based on TFF3-related risk scores,T stage,N stage,age and gender had good predictive value and can bring more net benefits to patients.Conclusion The prognostic model constructed based on TFF3-related genes can not only effectively predict the prognosis of patients,but also predict the efficacy of immunotherapy.关键词
