升麻素苷调控lncRNA CIR改善IL-1β诱导的软骨细胞外基质稳态失衡的研究

Abstract
Objective:Observe the effect of prim-O-glucosylcimifugin(POG)on cartilage extracellular matrix(ECM)homeostasis in IL-1β-induced mouse chondrocytes by regulating long non-coding RNA(lncRNA)CIR.Methods:C57BL/6 mouse knee cartilage was isolated and cultured in vitro after isoflurane anesthesia.The mouse chondrocytes were randomly divided into a blank group,a model group,and a POG group,and the corresponding interventions were administered.Real-time PCR was used to detect changes in CIR levels in mouse chondrocytes after POG intervention.Western blot analysis was used to detect the expression of MMP-9,MMP-13,ADAMTS-5,and caspase-3 proteins.Immunofluorescence was used to detect the expression of MMP-13 and caspase-3 in chondrocytes after cohogenin intervention.Results:Compared to the blank group,the levels of lncRNA CIR in the model group were significantly increased(P<0.05).Compared to the model group,the levels of lncRNA CIR in the POG group were significantly decreased(P<0.05).The expressions of MMP-9,MMP-13,ADAMTS-5,and caspase-3 proteins in the POG group were significantly decreased(P<0.05).The expressions of MMP-13 and caspase-3 fluorescence were significantly decreased after POG intervention.Conclusion:POG can improve the abnormal extracellular matrix homeostasis of IL-1β-induced chondrocytes,which is related to the regulation of lncRNA CIR,MMP-9,MMP-13,ADAMTS-5,and Caspase-3.关键词
