Abstract

Objective To investigate the role of interleukin-27(IL-27)in inducing macrophage polarisation towards the M2 type and thus exerting a therapeutic effect in acute pancreatitis(AP).Methods To compare the changes of inflammatory factors in AP mice.The cell model of acute pancreatitis was established by using INF-γ(500 U/mL)+LPS(100 ng/mL),which induced the polarisation of RAW264.7 macrophages to M1 type,and then co-culturing the RAW264.7 macrophages overexpressing IL-27 with them to observe whether the macrophages were polarised from M1 to M2 type,and to detect the related changes of inflammatory factors.Finally,the above related molecular mechanisms were preliminary explored by ITRAQ quantitative proteomics assay.Results In AP mice,IL-27 expression was reduced and other pro-inflammatory factors were significantly overexpressed.After transfection of RAW264.7 macrophages with IL-27,the cell viability of RAW264.7 macrophages was completely unaffected,the expression of the M1 marker CD86 was significantly decreased(P<0.01),and the expression of the M2 marker CD206 was significantly increased(P<0.05).M1 macrophages were significantly increased after local inflammatory stimulation,but the expression of CD86 was greatly reduced(P<0.05),and the expression of the inflammatory factor TNF-ɑ was also significantly decreased(P<0.01)after co-culture of macrophages transfected with IL-27.ITRAQ quantitative proteomics results indicated that the above effects may be related to the NF-κB and PI3K/AKT signaling pathways,and further Western blotting results were consistent with the proteomics results.Conclusion IL-27 induces the polarisation of RAW264.7 macrophages from M1-type to M2-type via the NF-ĸB and PI3K/AKT pathways,which reduces the inflammatory response and achieves the therapeutic effect in acute pancreatitis.
