臭椿酮通过HSP90/PKM2/STAT3信号通路抑制HCC细胞的活性

Abstract
Objective:To investigate the effect and potential mechanism of Ailanthone(AIL)in inducing human hepatocellu-lar carcinoma SMMC-7721 cell death.Methods:SMMC-7721 cells were cultured in vitro and the experiment was divided into the control group and the AIL administration group.Cell proliferation activity was detected by CCK-8 assay.Transwell invasion assay and cells scratch assay were used to detect the invasion and migration ability of cells.Lactic acid production and glucose content were detected by the kit.The apoptosis level was detected by flow cytometry.The expressions of HSP90/PKM2/STAT3 signal-ing pathway and apoptosis-related proteins Bax and Bcl-2 on SMMC-7721cells were detected with Western blot.The interaction between PKM2 and HSP90 and between PKM2 and STAT3 protein were detected by immunofluorescence.Results:AIL inhibit-ed the viability of SMMC-7721 cells,weakened the ability of cell invasion and migration,inhibited lactic acid production and in-creased glucose content(P<0.05).The results of flow cytometry showed that AIL induced SMMC-7721 cell apoptosis in a concentration-dependent manner,and the expression of apoptosis-related protein Bax increased,while the expression of Bcl-2 de-creased(P<0.05).In addition,AIL significantly inhibited the expression of PKM2 and STAT3(P<0.05),but had no signifi-cant effect on the inhibition of HSP90(P>0.05).The results of immunofluorescence double staining showed that AIL inhibited the formation of PKM2/HSP90 and PKM2/STAT3 complexes.Conclusion:AIL regulates cellular glycolysis through the HSP90/PKM2/STAT3 signaling pathway induces apoptosis in human hepatocellular carcinoma SMMC-7721 cells.关键词
