Abstract

Objective:To investigate the effective breast cancer sites of Cuscuta and study its mechanism of action.Methods:Extraction of Cuscuta with organic solvents of different polarity and the effective parts were screened under the guidance of pharma-cological effects.The effects of different extraction sites of Cuscuta on the proliferation of 5 tumor cell lines were assessed using MTT colorimetry,and the optimal extraction sites and tumor-sensitive strains were identified.The impact of a 95%alcohol ex-tract from Cuscuta on the morphological changes,migration,invasion,and apoptosis of MCF-7 cells was evaluated through invert-ed microscope observation,strip testing,Transwell assay,and Annexin V-FITC/PI double staining.Western blot was employed to analyze the expression levels of PI3K,p-PI3K,AKT,p-AKT,Bax,and Bcl-2 proteins in MCF-7 cells.Results:The prolifer-ation of MCF-7,SGC-7901,HepG-2,Hela and A549 cells was inhibited by all the extracted parts of Cuscuta.Among them,the inhibitory effect of chloroform and 95%alcohol extract from Cuscuta on the proliferation of MCF-7 cell line was better than the oth-er parts.The median inhibitory concentration(IC50)was 14.3 and 738.0 μg/mL,respectively.After treatment with 400,800,1 200 μg/mL of 95%alcohol extract of Cuscuta on MCF-7 cells,compared to the control group,the cells became round and con-stricted.The space between adjacent cells in the cell mass increased and the migration and invasion ability were gradually weak-ened.Additionally,there was an increase in apoptosis rate(P<0.05),in a time-and dose-dependent manner.Western blot results showed that Cuscuta's 95%alcohol extract could down-regulate the expression of p-PI3K,p-AKT,and Bcl-2 proteins in MCF-7 cells while up-regulating Bax protein expression as well as increasing the ratio of Bax/Bcl-2(P<0.05).Conclusion:Different ex-tracts of Cuscuta demonstrated significant inhibitory effects on the proliferation of 5 tumor cell lines.Notably,the chloroform and 95%alcohol extract of Cuscuta exhibited enhanced inhibitory effects on MCF-7 cells.The anti-breast cancer mechanism attributed to the 95%alcohol extract of Cuscuta may be linked to its modulation of the PI3K/AKT pathway,regulation of downstream Bcl-2 and Bax protein expressions,and participation in the mitochondrial apoptosis pathway.
