Abstract

Objective:To explore the key genes involved in airway remodeling in severe asthma(SA)based on bioinformatics and to map database-predicted traditional Chinese medicines,providing insights for clinical treatment of SA.Methods:SA-related datasets were ob-tained from the GEO database,and airway remodeling genes were retrieved from the Genecards database.Genes associated with SA airway remodeling were identified,and functional enrichment analysis was performed on these genes.Machine learning methods were then used to further screen for key differentially expressed genes in SA airway remodeling.ROC validation was conducted using both internal and exter-nal GEO datasets.Potential traditional Chinese medicines targeting SA airway remodeling were identified by mapping the key genes with the Coremine database.Results:This study identified a total of 53 differentially expressed genes in severe asthma(SA),including 29 up-regulated genes and 24 downregulated genes.Twelve genes related to airway remodeling in SA were obtained from which six characteristic genes were selected.A total of 120 traditional Chinese medicines(TCMs)were predicted to potentially act on SA airway remodeling,mainly including Heat-clearing drugs,tonifying drugs as well as Phlegm-resolving and cough-relieving bronchodilators.Molecular doc-king showed that the characteristic genes had good binding with the core components of high-frequency TCMs.Conclusion:Vascular en-dothelial growth factor A,fibronectin,CXADR,keratin,interleukin-1 receptor type 2,and interleukin-18 receptor 1 may be potential bi-omarkers for the treatment of airway remodeling in SA.It is speculated that TCMs such as Aidicha(Ardisia herb),Banzhilian(Scutellaria barbata),Nzhenzi(Ligustrum lucidum)and Baiguo(Ginkgo seed)may be potential therapeutic drugs for SA airway remodeling.
