摘要

Abstract

[Objective]To tackle sectioning challenges and background noise encountered in traditional detection of endophytic fungi in Kadsura coccinea(a plant characterized by highly lignified and deeply pigmented tissues),this study aims to develop a rapid and efficient aniline blue staining-micro-scopic detection method for endophytic fungi based on petal tissue.[Method]Petals of K.coccinea were used as experimental materials.Samples were prepared via hand-tearing and whole-mount methods,stained with varying concentrations of aniline blue solution,and their detection effects on endophytic fungi were systematically observed and compared.[Result]Endophytic fungi were successfully detected in K.coccinea petals,confirming the feasibility of using petals as detection materials.Notably,samples from the hand-tearing method exhibited clearer microscopic fields,better hyphal staining,and significantly higher contrast between hyphae and petal background than the whole-mount method.[Con-clusion]This study established an in situ detection technique for endophytic fungi in K.coccinea petals,verifying that the hand-tearing method yields superior observation results to the whole-mount method.With advantages of rapidity,clarity,and low cost,this technique provides reliable technical support for microecological research on K.coccinea and other medicinal plants with similar morphological characteris-tics.
