Abstract

Objective:To investigate the effect and mechanism of electroacupuncture in regulating neural plasticity in rats with cerebral infarction.Methods:Fifty-four SPF-grade male rats were selected as research subjects and randomly divided into the sham operation group,model group,and electroacupuncture group,with 18 rats in each group.The sham operation group only had the bilat-eral common carotid arteries exposed followed by incision suture.The model group and electroacupuncture group were established as rat models of middle cerebral artery occlusion(MCAO).After successfully modeling,the model group was only anesthetized and fixed,while the electroacupuncture group received electroacupuncture at bilateral Zusanli(ST36)and Neiguan(PC6)after anesthesia and fixation.Neurological function,cerebral infarction volume,the nerve cell morphology and synapses in the ischemic penumbra,the ex-pression of synapse-related proteins and Sonic Hedgehog(Shh)pathway proteins were compared among the three groups.Results:Compared with the sham operation group,rats in the model group exhibited large-area cerebral ischemic lesions,with a reduced num-ber and abnormal morphology of nerve cells,as well as abnormal synaptic morphology in the ischemic penumbra.Compared with the model group,rats in the electroacupuncture group showed a decreased area of cerebral ischemic lesions(P<0.05),an increased num-ber of nerve cells,a reduced number of degenerated neurons,and improved synaptic morphology in the ischemic penumbra.Additionally,the expressions of Shh,Smoothened(Smo),Glioma-associated oncogene homolog 1(Gli1),and Synaptophysin were significantly increased(P<0.05),while the expression of Postsynaptic Density Protein 95(PSD-95)showed no significant change.Conclusion:Electroacupuncture improves neurological function and promotes nerve cell growth in cerebral infarction rats,possibly via activating the Shh pathway to enhance neural plasticity.
