Cloning of the Peppers Transcription Factor Gene CaNAC100 and Its Expression in Response to Abiotic Stress and Hormone Treatments

Abstract
[Objective]This study aims to investigate the expression patterns of the pepper transcrip-tion factor CaNAC100 under abiotic stress and hormone treatments,thereby laying a foundation for further elucidating its molecular mechanism in regulating stress resistance in pepper.[Method]The CDS sequence of CaNAC100 gene was obtained by homologous cloning method using Yunnan local yellow pep-per,wrinkled yellow pepper and purple pepper as experimental materials,and its coding protein and cis-acting elements in promoter region were analyzed by bioinformatics.The tissue-specific expression pattern of CaNAC100 gene and its expression level in roots under abiotic stress and hormone treatment were quan-titatively analyzed by RT-qPCR.[Result]The CDS sequence of the CaNAC100 gene was 954 bp in length,encoding 317 amino acids.CaNAC100 protein is a hydrophilic unstable protein with a molecular weight of 36.12 kD and which is localized in the nucleus.Phylogenetic analysis showed that the CaNAC100 protein cloned in this study was closely related to the NAC100 protein of Capsicum baccatum and Capsicum chinense.Phylogenetic tree analysis indicated that the CaNAC100 protein cloned in this study was most closely related to that of Capsicum baccatum and Capsicum chinense.The expression pat-tern analysis showed that CaNAC100 had the highest expression level in flowers,followed by roots and leaves.Stress and hormone response analysis showed that CaNAC100 had different degrees of response to different stresses and hormone treatments.It had the slowest response to salt stress and drought stress,the fastest response to low temperature,followed by high temperature and methyl jasmonate(MJ).At 24 h of stress exposure,the highest response intensity was observed to abscisic acid(ABA)treatment,fol-lowed by low temperature,high salt stress,and drought stress.[Conclusion]The CaNAC100 gene of pep-per was mainly involved in the response to ABA treatment,and also participated in the response to low temperature,high salt stress,and drought stresses.These findings provide a basis for further research on the biological functions and molecular mechanisms of stress resistance mediated by CaNAC100 gene.关键词
