新铁炮百合离体再生体系的优化研究
Optimization of the in vitro Regeneration System for Lilium×formolongi

Abstract
[Objective]It is to establish and optimize the in vitro regeneration and propagation system for Lilium×formolongi scales.[Method]In this paper,scales of Lilium×formolongi were used as explants.The optimal type of the best antibacterial agent was screened.Through orthogonal experimental design,the effects of different types and concentrations of hormones on the induction of adventitious buds and bulb enlargement and rooting in lily scales were investigated.[Result]The optimal antibacterial agent for tissue culture of Lilium×formolongi scales was PPM,a plant tissue culture antibacterial agent.The scale contamination rate was 15%,the adventitious bud regeneration rate was 75%,and the average number of adventitious buds regenerated was 2.1.The optimal medium formulation for adventitious bud induction of Lilium×formolongi scales was MS+1.0 mg/L 6-BA+0.5 mg/L CTK+1.0 mg/L NAA+30 g/L sucrose+7.5 g/L agar,with an adventitious bud induction rate of 93.07%;The optimal bulb enlargement culture medium formulation was MS+1.0 mg/L 6-BA+5.0 mg/L PP333+2.0 mg/L IAA+80 g/L sucrose+7.5 g/L agar,with a bulb enlargement rate of up to 50.24%after 30 days.[Conclusion]Through screening for the best antibacterial agents and optimizing hormone ratios,this study established an effi-cient in vitro regeneration and propagation system for Lilium×formolongi scales,providing the material and technical foundation for future genetic transformation regeneration experiments.关键词
