Abstract

This study aims to enhance the antioxidant activity and bioavailability of Coenzyme Q10(CoQ10)by preparing CoQ10 nanoemulsion using medium chain triglycerude as the oil phase,soybean phospholipid as the surfactant,and Tween-80 as the co-surfactant.The preparation process of CoQ10 nanoemulsion was optimized using Box-Behnken response surface methodology.Also,the physicochemical properties,stability,antioxidant activity,and bioavailability of the CoQ10 nanoemulsion were investiga-ted.The results show that the optimal conditions for CoQ10 nanoemulsion were 2 g of medium chain trig-lycerude,5.353 g of soybean phospholipid,and 1 g of Tween-80.In this case,the average particle size,polydispersity index,zeta potential,and encapsulation efficiency of the CoQ10 nanoemulsion were(72.9±0.32)nm,0.25±0.034,(-34.9±1.97)mV,and(97.6±0.49)%,respectively.Transform infrared spectroscopy results show that CoQ10 was completely encapsulated in the nanoemulsion.Moreover,the result of Transmission Electron Microscopy indicates that the CoQ10 nanoemulsion were round,non-ag-gregated,and evenly dispersed in the system.The stability experiments demonstrate that CoQ10 nanoe-mulsion displayed better centrifugal stability,ionic stability,and storage stability.Additionally,com-pared to CoQ10 suspensions,the DPPH free radical,ABTS free radical,and hydroxyl free radical scaven-ging rates of CoQ10 nanoemulsion was increased by 79.1%,93.1%and 72.2%,respectively.The blood concentration of CoQ10 in rats for the optimized CoQ10 nanoemulsion was 0.767 Pg/mL,which was 1.405 times that of CoQ10 oil.This study could provide data support for the development of Coenzyme CoQ10 nanoemulsion,and lay the foundation for its application in health food and biomedicine fields.
