Abstract

In order to add new content to the study of seed aging theory,DNA methylation variation of aged maize seeds was studied.In the trial,aged seeds of six maize inbred lines were selected as ex-perimental materials in comparison with new seeds of the same set of lines.DNA methylation varia-tion at the 5′-CCGG sites was analyzed by the MSAP marker.The results showed that aged maize in-bred line seeds have undergone varying degrees of changes in DNA methylation levels and patterns,and the levels of DNA methylation at 5′-CCGG sites of the aged maize inbred lines either increased or decreased,with the respective percentage ranging from 0.24%to 4.03%for the increase and from 1.05%to 1.22%for the decrease,respectively.Three types of DNA methylation variation patterns were identified in the aged seeds.The total percentage of the four patterns of single-strand CHG hemimethylation variation ranged from 5.45%to 10.42%.The percentage of the four patterns of double-strand CG full methylation variation in the DNA of the aged seeds ranged from 4.26%to 10.29%.The total percentage of simultaneous variation in CG and CHG methylation in DNA of the aged seeds ranged from 3.66%to 5.69%.According to χ2 test,there was an extremely significant cor-relation between DNA methylation variation pattern and different maize inbred lines,among which the percentage of DNA methylation variation in the aged seeds of inbred line PHJEV was the highest,reaching 77.24%.
