Effect of spinal cord injury on urodynamics in rats and its mechanism
摘要
Abstract
Objective To investigate the effects of spinal cord injury on urodynamics and the endothelin-1(ET-1)levels in the blood and urine of rats. Methods A total of 24 female Wistar rats were subjected to bladder catheterization and randomly divided into experimental group and control group, with 12 rats in each group. All rats were kept under identical conditions. The experimental rats received a spinal cord transection to establish a spinal cord injury model, while the control rats were not subjected to this procedure.Urodynamic tests were performed at 2 and 6 weeks post-modeling.ET-1 levels in plasma and urine were mea-sured using ELISA at 2,4,and 6 weeks post-modeling.The measured parameters were compared between the two groups.Re-sults At 2 weeks post-modeling,the experimental group had significantly higher bladder capacity and compliance(Fintergroup=414.809,6.481,P<0.05)and significantly lower detrusor pressure(Fintergroup=989.697,P<0.001)compared to the control group. At 6 weeks post-modeling,the experimental group had significantly lower bladder capacity and compliance(Fintergroup=37.177,247.380,P<0.001)and significantly higher detrusor pressure(Fintergroup=161.798,P<0.001)compared to the control group. In the experimental group, the bladder capacity and compliance were significantly higher at 2 weeks post-modeling than those before modeling, and significantly lower at 6 weeks post-modeling than those before modeling and 2 weeks post-modeling(Fintragroup=579.401,7.850,P<0.05).In the experimental group, the detrusor pressure was significantly lower at 2 weeks post-modeling than that before modeling, and significantly higher at 6 weeks post-modeling than that before modeling and 2 weeks post-modeling(Fintragroup=831.148,P<0.001).Furthermore,the levels of urinary ET-1 were significantly lower in the experimental group than in the control group at 2,4 and 6 weeks post-modeling(Fintragroup=493.067-1 622.096,P<0.001).The levels of urinary ET-1 af-ter modeling decreased in the experimental group(Fintragroup=233.323,P<0.001)and showed no significant changes in the control group(P>0.05).Conclusion During the spinal shock phase following spinal cord injury in rats, the bladder capacity and com-pliance increased while detrusor pressure decreased. After the shock phase, the bladder capacity and compliance decreased and the detrusor pressure increased. The mechanism underlying these changes may be related to the decrease in urinary ET-1 levels.关键词
