Abstract

Alkaline hydrogen peroxide(AHP)pretreatment was used to pretreat bamboo(Bambusa emeiensis),and the recovered lignin was obtained from the AHP pretreatment liquor by acid precipitation,realizing the mild and effective lignin separation.The structural properties of recovered lignin(L0-L3.0)were investigated with Fourier transform infrared spectroscopy(FT-IR),gel permeation chromatography(GPC),two-dimensional heteronuclear single-quantum coherence nuclear magnetic resonance(2D-HSQC NMR)and thermogravimetric analysis(TG).Moreover,the antioxidant activity of recovered lignin was also evaluated.The results showed that the AHP pretreatment could remove lignin effectively,the maximum of lignin removal yield was 70.97%at 3.0%H2O2 dosage,and the yield of lignin samples was up to 60.18%at 0%H2O2.The 2D-HSQC NMR spectra showed that the structure of removed lignin was well preserved,which mainly composed of syringyl(S)and guaiacyl units(G),and the values of S/G were 1.45-1.50.In addition,a predominance of β-O-4·linkages up to 54.76%-59.84%,the contents of β-β·(B)and β-5·(C)linkages were 3.04%-4.10%and 1.32%-2.73%,respectively.Gel permeation chromatography showed that compared to cellulolytic enzyme lignin(CEL),the relative molecular mass of removed lignin was lower,and the polydispersity index(PDI)was stable at around4.80.Furthermore,as the dosage of H2O2 increased from 0%to 2.0%,the removed lignin exhibited favorable thermal stability and antioxidant activity.Most of them degraded in the temperature range of 250-400℃,and the half maximal inhibitory concentrations(IC50)values of DPPH free radical scavenging ability were 17.62(L0),16.26(L1.0),and15.99(L2.0)mg/L,respectively,which were lower than28.97 mg/L of CEL.
