Abstract

Using choline chloride(ChCl)as a hydrogen bond acceptor,thirty-six deep eutectic solvent(DES)were synthesized and compared with the conventional solvent ethanol(EtOH)for the extraction of total flavonoids(TF)from Lycium barbarum pomace.The optimal DES system was determined based on extraction yield,and the ultrasound-assisted extraction conditions for TF were optimized using single-factor experiments and response surface methodology.The flavonoid compounds in L.barbarum pomace were quantified using liquid chromatography-tandem mass spectrometry(LC-MS/MS),and the antioxidant activity of the extracts was evaluated.The results indicate that using a DES composed of choline chloride and 1,2-butanediol/triethylene glycol at a molar ratio of 1∶1.35 as the extraction solvent,with14%water content of DES,a liquid-to-material ratio of 24∶1(mL∶g),an ultrasonic extraction temperature of 45℃,and an ultrasonic extraction time of 49 min,achieved a TF yield of 13.104 mg/g.The yield was significantly higher than the3.272 mg/g,which was obtained through traditional ethanol extraction.Five flavonoids of kaempferol,chrysin,spiraeoside,tiliroside,and phloretin were identified in the crude extract of DES.None of them was detected in the crude extract of 60%ethanol.Notably,the contents of isorhamnetin and apigenin were found to be 4.21 and 3.88 times higher,respectively,compared to those in the crude extract of 60%ethanol.Furthermore,within a certain mass concentration range,the scavenging rate of superoxide anions and hydroxyl radicals as well as the total antioxidant capacity of the DES extract generally surpasses that of the 60%ethanol extract.
