慈竹秆多糖加压热水提取工艺优化及其抗氧化活性评价
Optimization of Pressurized Hot Water Extraction of Bambusa emeiensis Culm Polysaccharides and Evaluation of Their Antioxidant Activity

Abstract
Polysaccharides were extracted from the culms of Bambusa emeiensis L.C.Chia&H.L.Fung using a pressurized hot water extraction(PHWE)method.The extraction conditions were optimized by response surface methodology,the antioxidant activity of the polysaccharides was evaluated by free radical scavenging test,and the structure of the purified polysaccharide was analyzed by gel permeation chromatography(GPC)and infrared spectroscopy.The results indicated that extraction temperature significantly affected the total sugar yield,while extraction time significantly influenced the degree of polysaccharide degradation(polysaccharide/total sugar ratio).The optimal extraction conditions were determined as follows:extraction temperature of 139.5℃,extraction time of 32 min,and a liquid-to-solid ratio of 15.5∶1(mL∶g).Under these conditions,the yield of total sugars from B.emeiensis culms was 1.39%,the polysaccharide yield was 0.81%,and the polysaccharide/total sugar ratio was 0.57.The polysaccharides exhibited considerable antioxidant activity,with half-maximal inhibitory mass concentration(IC50)values for scavenging DPPH·,·ABTS+,and·OH being 1.89,0.86,and 0.22 g/L,respectively.The polysaccharides were separated and purified using a DEAE-52 of cellulose column,yielding one purified polysaccharide fraction,designated PWDP-N.PWDP-N had a weight-average molecular mass of 14 484 u and a dispersity index of 1.00.It exhibited characteristic infrared absorption peaks of a neutral polysaccharide.Monosaccharide composition analysis revealed that PWDP-N was composed of eight monosaccharides,including mannose,galactose,and glucose.关键词
