Exploring the Pathogenesis of Osteoarthritis by Combining GEO with Mendelian Randomization

Abstract
Objective To explore the pathogenesis of osteoarthritis(OA)by integrating Gene Expression Omnibus(GEO)-database analysis with Mendelian randomization.Methods The chip containing OA and synovial membrane gene data of healthy people was downloaded from GEO.After classification into control and test groups,differential genes were identified with limma and weighted gene co-expression network analysis(WGCNA).Core genes were then selected via the STRING platform and Cytoscape software.Among them,MYC proto-oncogene proteins related to core genes and with research value were screened from the whole genome sequencing data(GWAS)database as exposure data.SNPs strongly associated with MYC were used as instrumental variables,and Mendelian randomization analysis was conducted using knee OA(KOA),OA,and coxitis(COX)as outcomes data respectively.Results After screening,it was finally determined that IL1B,JUN,MYC,CD4 and PTPRC were the top five genes in terms of the number of gene interaction nodes.The MYC proto-oncogene protein and KOA were analyzed by Mendelian randomization.The OR value detected by the inverse variance weighted method(IVW method)was 1.103(95%CI:1.002-1.215),indicating a positive correlation between the two(P value=0.046),indicating that there might be a causal relationship between them.Conclusion The MYC proto-oncogene protein may be a potential risk factor for the onset of KOA.关键词
