甘西鼠尾草中丹参酮类成分的分离鉴定及其治疗溃疡性结肠炎的机制
Isolation and identification of tanshinones from Salvia przewalskii Maxim.and their mechanisms against ulcerative colitis

Abstract
Objective To isolate and identify tanshinones from the dried whole plant of Salvia przewalskii Maxim.and investigate their mechanisms against ulcerative colitis using network pharmacology and molecular docking techniques.Methods The tanshinones from the dried whole plant of Salvia przewalskii Maxim.were isolated using separation techniques such as silica gel column chromato-graphy,ODS reverse-phase chromatography,and prep-HPLC.The structures were identified through spectroscopic methods,including ¹H-NMR and ¹³C-NMR.The potential target proteins of the isolated tanshinones were predicted based on the Swiss Target Prediction database.The targets of ulcerative colitis were retrieved from the GeneCards and OMIM databases.The"drug-ingredient-target-disease"network and PPI core target network were constructed respectively.The GO function enrichment analysis and KEGG pathway analysis were performed on the core targets,and molecular docking was performed using AutoDock.Results Three tanshinones were isolated from the dried whole plant of Salvia przewalskii Maxim.Structural identification confirmed that they are all abietane-type diterpenes,namely tanshinone ⅡA,cryptotanshinone and tanshinone Ⅰ.A total of 134 potential targets of tanshinones,5 335 ulcerative colitis targets and 82 intersecting targets were screened.The PPI analysis identified 18 key targets.The GO enrichment analysis of the potential targets of tanshinones in the treatment of ulcerative colitis primarily involved in inflammatory response.The KEGG pathway analysis mainly involved in Toll-like receptor signaling pathway and NF-κB signaling pathway.The molecular docking showed that three tanshi-nones had good binding ability to IL-6,TNF and IL-1β.Conclusion The tanshinones isolated from the dried whole plant of Salvia przewalskii Maxim.are the abietane-type diterpenoids,and the tanshinones may exert anti-ulcerative colitis effects by regulating the Toll-like receptor signaling pathway and the NF-κB signaling pathway through IL-6,TNF and IL-1β.关键词
