白凤菜乙醇提取物对大鼠高尿酸血症及痛风性肾病的影响
Effects of the ethanol extract from Gynura formosana Kitam on hyperuricemia and gouty nephropathy in rats

Abstract
Objective:To investigate the effects of the ethanol extract of Gynura formosana Kitam on a rat model of hy-peruricemia induced by potassium oxonate combined with adenine.Methods:Sixty male SD rats were randomly divided into 6 groups(n=10 per group):blank control group,model group,allopurinol group(30 mg/kg),high-dose(10.0 mg/ml),medium-dose(5.0 mg/ml),and low-dose(2.5 mg/ml)groups of ethanol extract from Gynura formosana Kitam.The hyper-uricemia model was established using potassium oxonate combined with adenine for 28 consecutive days.Starting on the 8th day of modeling,rats in the blank and model groups were administered a 0.5%sodium carboxymethylcellulose(0.5%CMC-Na)solution daily in the afternoon.Rats in each treatment group received their respective drugs at a dose of 1 ml/100 g for 21 consecutive days.Blood samples were collected on days 0,14,and 28 to measure serum uric acid(UA),serum creatinine(Cr),and blood urea nitrogen(BUN)levels.On days 14 and 28,concentration of xanthine oxidase(XOD)in serum was de-termined.At the end of the experiment,liver tissues were homogenized with 4 times the volume of normal saline and centri-fuged,and the supernatant was used to measure liver XOD content.To observe pathological changes in the kidney,the right kidney was collected for hematoxylin-eosin(HE)staining and pathological sectioning.Results:On both day 14 and day 28,levels of serum Cr,UA,and blood urea nitrogen(BUN)in rats treated with high-dose Gynura formosana Kitam ethanol ex-tract,as well as those treated with allopurinol,were all significantly lower than those in the model group(P<0.01).On day 14,BUN levels in the high-and medium-dose Gynura formosana Kitam ethanol extract groups were significantly lower than those in the allopurinol group;on day 28,BUN levels in the high-dose Gynura formosana Kitam ethanol extract group re-mained significantly lower than those in the allopurinol group(all P<0.01).On both day 14 and day 28,serum UA levels in all dose groups of Gynura formosana Kitam ethanol extract were significantly higher than those in the allopurinol group(P<0.01).On both day 14 and day 28,there were no statistically significant differences in serum Cr levels between any dose group of Gynura formosana Kitam ethanol extract and the allopurinol group(P>0.05).Similarly,serum and hepatic xan-thine oxidase(XOD)levels in the high-dose Gynura formosana Kitam ethanol extract group showed no significant differ-ence compared with the allopurinol group(P>0.05).These findings suggest that,compared to allopurinol,Gynura formo-sana Kitam extract at all tested doses was less effective in lowering serum UA;however,the high and medium doses dem-onstrated superior efficacy in reducing BUN.Regarding serum Cr reduction,all doses of Gynura formosana Kitam ethanol extract performed comparably to allopurinol.Moreover,the high-dose Gynura formosana Kitam ethanol extract exhibited a similar effect to allopurinol in reducing both serum and hepatic XOD activity.Histopathological examination revealed that,compared with the model group,rats in the high-dose Gynura formosana Kitam ethanol extract group showed no obvious changes or only mild alterations in glomerular volume,Bowman's capsule space,and renal tubular luminal diameter.Addi-tionally,inflammatory cell infiltration around the glomeruli and renal tubules was markedly reduced,and tubular epithelial cells exhibited no significant edema.These observations indicate that Gynura formosana Kitam ethanol extract exhibits no apparent nephrotoxicity in hyperuricemic rats.Conclusion:The ethanol extract of Gynura formosana Kitam can effectively reduce levels of serum UA,Cr,and BUN in a rat model of hyperuricemia induced by potassium oxonate combined with ad-enine.This therapeutic effect may be associated with its ability to decrease hepatic xanthine oxidase(XOD)activity.关键词
