Abstract

In this study,the F1 generation of Aohu sheep population was used as the research subject.PCR sequencing was employed to detect three single nucleotide polymorphism(SNP)sites on Exon 20 of the RASA2 gene in 600 ewes.Using PASW Statistics 18.0 GLM and SPSS 22.0,the association between mutations at the three SNPs of the RASA2 gene(g.245315235T>A,g.2451314883T>C,and g.2451314934T>C)and the litter sizes of ewes aged 2-3 years with different body weights(41-50 kg,51-60 kg,61-70 kg)across different parities was analyzed.The results showed that three SNPs loci were detected in the RASA2 gene:g.245315235T>A,g.2451314934T>C,and g.2451314883T>C loci,which yielded the dominant genotypes:TA,TC,and TC,respectively,and all of them were moderately polymorphic,among which,g.245315235T>A belonged to a missense mutation at codon 422 position 3,resulting in the mutation of amino acid from cysteine(Cys)to glutamic acid(Glu),g.245314883T>C belongs to a missense mutation,at codon 366 position 2,resulting in the change of amino acid from methionine(Met)to Cys,g.245314934T>C belongs to a missense mutation,at codon 320 position 2,leucine(Leu)mutated to proline(Pro).all three SNPs loci were in Hardy-Weinberg equilibrium(P>0.05),but there was linkage disequilibrium between the three SNPs two by two,yielding four haplotypes,TA,TC,CA,and CC.association analysis revealed that TA,TC,and TC mutant genotypes with body weights ranging from 51-60 kg were significantly correlated with the number of lambs produced in different litters.The aggregated genotypes were mainly dominated by TTAA and TCTC.The present study revealed that polymorphisms at the SNPs locus of the RASA2 gene are strongly permeable in the evolutionary selection process of sheep reproductive traits,which may provide a referable data basis for the breeding and selection of new breeds(lines)of meat sheep with multiple births in the future.
