Abstract

Objective To establish a reliable method for isolating and purifying primary liver cells from mice with high vitality and purity of liver fibrosis.Methods The classical two-step collagenase perfusion method was improved by using in-ferior vena cava perfusion,intermittent clamping of the portal vein perfusion,and 50%Percoll density-gradient centrifugation for purification of hepatocyte suspension.The purified hepatocytes were cultured in vitro,and immunofluorescent staining of Albumin and HNF1α was performed.Results The survival rate of primary liver cells isolated and purified from liver fibrosis mice could stably reach(92.93±1.93)%,with an average total number of live cells of(1-2)×107 per adult mouse.Purified cells adhered to the substrate and began to spread within 2 h of in vitro culture.Conclusion A stable tech-nique for isolating and culturing primary liver cells from mice with high purity and vitality in liver fibrosis is successfully established.
