Correlation between the platelet-derived growth factor-BB/platelet-derived growth factor receptor-β signaling pathway and the progression and prognosis of diabetic retinopathy

Abstract
Objective To analyze the levels of platelet-derived growth factor-BB(PDGF-BB)and platelet-derived growth factor receptor-β(PDGFR-β)in patients with diabetic retinopathy(DR)and investigate their correlation with the progression and prognosis of DR.Methods A total of 116 patients(116 eyes)with DR in the Affiliated Hospital of Shan-dong University of Traditional Chinese Medicine from May 2022 to May 2025 were selected as the DR group,and another 116 patients(116 eyes)with diabetes mellitus(DM)without DR during the same period were selected as the non-DR(NDR)group.The levels of PDGF-BB and PDGFR-β in aqueous humor and serum were compared between the DR group and the NDR group,as well as among the patients with different disease severities in the DR group,and the correlation between PDGF-BB/PDGFR-β levels and DR progression was analyzed.The DR group was followed up for 1 year.The clinical data,as well as the levels of PDGF-BB and PDGFR-β in aqueous humor and serum,were compared between patients with different prognostic outcomes.Logistic analysis was used to analyze the relationship between each index and prognosis.The receiver operating characteristic(ROC)curves were drawn to analyze the predictive value of PDGF-BB and PDGFR-β levels in aque-ous humor and serum for the prognosis of patients.Results The levels of PDGF-BB and PDGFR-β in aqueous humor and serum of patients in the DR Group were higher than those in the NDR group(both P<0.05).The levels of PDGF-BB and PDGFR-β in aqueous humor and serum of patients with proliferative DR in the DR group were higher than those of patients with non-proliferative DR(both P<0.05).The levels of PDGF-BB and PDGFR-β in aqueous humor and serum were both positively correlated with the progression of DR(both P<0.05).The course of DM in patients with visual disability was longer than that in patients without visual disability,and their levels of HbAlc,as well as PDGF-BB and PDGFR-β in aqueous humor and serum,were all higher than those of patients without visual disability(all P<0.05).The levels of PDGF-BB and PDGFR-β in aqueous humor and serum were independently correlated with prognosis(both P<0.05).The areas under the curve(AUC)of PDGF-BB and PDGFR-β levels in aqueous humor and serum for predicting visual impairment in DR patients were 0.811,0.812,0.754,and 0.759.Conclusion PDGF-BB and PDGFR-β are highly expressed in the serum and aqueous humor of DR patients and are closely correlated with the progression and prognosis of patients.PDGF-BB and PDGFR-β in serum and aqueous humor have high predictive value for the prognosis of patients.关键词
