Abstract

To establish an efficient embryogenic callus-induced regeneration system for blueberry,this study inves-tigated the effects of different hormones,concentrations,dark culture time,leaf placement,and leaf parts on em-bryogenic callus induction and adventitious bud regeneration,using in vitro-grown'Legacy'blueberry leaves as ex-plants.The induction rates under different treatments were compared through visual analysis and multivariate analy-sis of variance.The results showed that the optimal induction medium was modified WPM+ZT 3.0 mg/L+IBA 0.1 mg/L medium(pH 5.2),combined with 14 days of dark cultivation.Using this medium,the embryogenic callus induction rate reached 100%,with an average of 9.12 regenerated buds per explant.Among the factors tested,ZT concentration had the most significant effect on embryogenic callus induction.Dark treatment for 14 days maximized the cell division capacity of embryogenic callus,enhancing adventitious bud induction and regeneration.Leaf polar-ity tests revealed that placing leaves with abaxial side upward yield higher regeneration efficiency than the adaxial side upward.Additionally,the petiole tip showed higher embryogenic callus induction compared to the leaf apex,with the proximal 1/3 of the petiole being the most effective induction site.
