摘要

Abstract

Objective To screen the better dose of 2,4,6-trinitrobenzene sulfonic acid(TNBS)for establishing ulcerative colitis(UC)model by comparing the efficacy of different TNBS-induced models.Methods SD rats were randomly divided into normal group,TNBS-50 mg/kg group,and TNBS-100 mg/kg group(n=18 in each group,half male and half female).The rats were fasted for 12 h before modeling.After anesthesia,the rats in TNBS groups were given 0.4 mL and 0.8 mL of 5%TNBS-ethanol solution by enema respectively,while the rats in normal group were given 0.8 mL of normal saline by enema.Body weight and fecal occult blood were monitored daily for disease activity index(DAI)scoring.Six rats in each group were sacrificed at days 1,7,and 14 after modeling,and then serum D-lactic acid and fecal albumin were detected,the colon length was measured,spleen and thymus were weighed,and colon tissues were stained with HE and AB-PAS.Results Compared with normal group,the rats in TNBS groups showed a significant decrease in body mass and an increase in DAI score from day 1 to day 4 of modeling;after day 4,the body mass loss slowed down,the mucopu-rulent bloody stool was alleviated,and the DAI score decreased;the mortality rates in TNBS-50 mg/kg group and TNBS-100 mg/kg group within 14 d were 5.6%and 27.8%,respectively.At day 1,the rats in both TNBS groups showed colon shortening(P<0.05),increased spleen coefficient(P<0.05),and elevated serum D-lactic acid and fecal albumin(P<0.05);at day 7,the rats in 50 mg/kg group exhibited colon shortening and elevated serum fecal albumin(both P<0.05),while the rats in both 50 mg/kg group and 100 mg/kg group showed decreased thymus coefficient and elevated serum D-lactic acid(both P<0.05);at day 14,the above indicators in 50 mg/kg group were still abnormal(P<0.05),while except for the significant increase of D-lactic acid,there was no difference in the other indexes between 100 mg/kg group and normal group.Pathohistological results showed that at day 1,obvious inflammation areas were seen in the colon in both TNBS groups,accompanied by submucosal edema,partial destruction of goblet cells and mucus layer injury;at day 7,the inflammation further aggravated,the epithelial cells were replaced by inflammatory cells,the inflammatory cells infiltrated into the muscular layer,and the goblet cells almost disappeared;at day 14,the lesion areas in both TNBS groups showed a recovery trend,but the recovery in 100 mg/kg group was more significant.Conclusion The 5%TNBS-ethanol solution at a dose of 50 mg/kg has significant advantages to induce the UC model,including low mortality rate,obvious and long-lasting colitis symptoms and patho-logical changes,and it serves as an optimal modeling condition for establishing the UC model.
