XIST和TSIX lncRNA在缺血性脑卒中中的作用及其机制
Role and mechanism of XIST and TSIX lncRNA in ischemic stroke

Abstract
Objective To identify potential lncRNA biological targets in ischemic stroke by performing RNA-seq on peripheral blood mononuclear cells(PBMCs)obtained from affected patients.Methods Fresh whole blood samples were collected separately from 10 ischemic stroke patients(case group)and 10 healthy controls(control group)using anticoagulant tubes.RNA was extracted from periphe-ral blood mononuclear cells(PBMCs).The transcripts were processed,assembled,and filtered using StringTie.Differentially ex-pressed long non-coding RNAs(lncRNAs)were identified using the DESeq2 package.The expression levels of the target mRNAs of these lncRNAs in PBMCs of ischemic stroke patients were validated by quantitative real-time reverse transcription polymerase chain re-action(qRT-PCR).Potential pathways involved in the trans-regulation and cis-regulation of the identified lncRNAs were explored us-ing Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis.Results By screening of candidate lncRNAs,a total of 1 391 expressed transcripts were identified in case group and control group.A total of 408 significantly differentially expressed lncRNAs were identified using DESeq2,including 262 up-regulated and 146 down-regulated lncRNAs.In-depth analysis of sequenc-ing results showed XIST and TSIX lncRNAs were potential novel biological targets in ischemic stroke.Compared to control group,the level of XIST mRNA expression was decreased in case group,while the level of TSIX mRNA expression was increased(both P<0.05).Co-expression analysis of the differentially expressed lncRNAs and mRNAs further revealed that the trans-regulated lncRNAs were primarily enriched in the molecular function of protein binding and centrally involved in the biological process of cell adhesion,and functions closely associated with extracellular vesicles.The cis-regulated lncRNAs were also mainly associated with protein binding,centered on immune responses as their core biological process,and were predominantly localized to the cell membrane.KEGG pathway enrichment analysis indicated that both regulatory modes were significantly enriched in the extracellular matrix(ECM)-receptor inter-action pathway.Conclusion XIST and TSIX lncRNAs can be potential biomarkers for the clinical diagnosis of ischemic stroke.They can induce the ischemic stroke by regulating the ECM-receptor interaction pathway.关键词
