大柴胡汤对动脉粥样硬化大鼠氧化三甲胺及脂代谢的影响
Effects of Da-Chai-Hu decoction on trimethylamine oxide and lipid metabolism in atherosclerotic rats

Abstract
Objective To investigate the regulatory effect of Da-Chai-Hu decoction on trimethylamine oxide and lipid metabolism in atherosclerotic model rats,and explore its potential anti-atherosclerotic mechanism.Methods SD rats were randomly divided into four groups using a random number table:normal control group,AS model group,Da-Chai-Hu decoction group,and Atorvastatin group,with 10 rats in each group.The concentrations of total cholesterol(TC),triglycerides(TG),low-density lipoprotein cholesterol(LDL-C),and high-density lipoprotein cholesterol(HDL-C)in serum were measured using a fully automated biochemical analyzer with matched reagent kits.Pathological changes of the aorta were observed by hematoxylin-eosin(HE)staining and oil red O staining.The serum TMAO concentration was measured using enzyme-linked immunosorbent assay(ELISA).Results Compared with normal control group,the body weight of rats in the other three groups increased significantly(P<0.05).Compared with AS model group,the body weight of rats in Da-Chai-Hu decoction group and Atorvastatin group decreased significantly(P<0.05).Compared with normal control group,the levels of TG,TC,and LDL-C increased,while HDL-C decreased in AS model group(all P<0.05).Compared with AS model group,the levels of TG,TC,and LDL-C decreased while HDL-C increased in Da-Chai-Hu decoction group and Atorvastatin group(all P<0.05).Compared with Atorvastatin group,the levels of TG,TC,and LDL-C increased while HDL-C decreased in Da-Chai-Hu decoction group(all P<0.05).HE staining results showed that the aortic structure was intact in normal control group,with smooth and continuous endothelium.The rats in model group exhibited severe aortic injury,characterized by significantly thickened intima,disordered medial fibers,and marked structural destruction.The degree of injury was alleviated in both Da-Chai-Hu decoction group and Atorvastatin group,showing a relatively flat intima,improved medial fibrous hyperplasia,and a more regular structure.Oil red O staining results showed almost no lipid deposition in normal control group.The rats in model group exhibited extensive red lipid deposition throughout the aortic wall.The red lipid deposition was significantly reduced in both Da-Chai-Hu decoction group and Ator-vastatin group,indicating reduced atherosclerotic plaque formation.Compared with normal control group,the serum TMAO concentration increased significantly in AS model group(P<0.05).Compared with AS model group,the TMAO concentration decreased signifi-cantly in both Da-Chai-Hu decoction group and Atorvastatin group(all P<0.05).Compared with atorvastatin group,the TMAO concentration significantly increased in Da-Chai-Hu decoction group(P<0.05).Conclusion Da-Chai-Hu decoction may exert anti-atherosclerotic effects by reducing TMAO concentration and regulating lipid metabolism disorders,thereby ameliorating abnormal body weight,dyslipidemia,and aortic pathological damage in atherosclerotic model rats.关键词
