EGCG对铅暴露小鼠致海马CA1区c-fos神经元损伤的保护作用
Protective effect of epigallocatechin gallate against c-fos neuron injury in hippocampal CA1 region in lead-exposed mice

Abstract
Objective To investigate the protective effect of epigallocatechin gallate(EGCG)against c-fos-positive neuronal damage in the hippocampal CA1 region of lead-exposed mice.Methods Eighty SPF-grade male C57BL/6 mice were randomly divided into four groups:control group,lead exposure group,EGCG intervention group,and EGCG+lead exposure group.Mice in control group and EGCG intervention group were given deionized water for drinking,while those in lead exposure group and EGCG+lead exposure group received drinking water containing 0.2%lead acetate.Starting from the 4th week of modeling,the mice in EGCG intervention group and EGCG+lead exposure group were subjected to intragastric administration of EGCG at a dose of 2 mg/kg body weight,three times a week for 4 consecutive weeks.The Morris water maze test and the novel object recognition test were used to evaluate the learning and memory abilities of mice.Western blot analysis was performed to detect the expression level of c-fos protein in the hippocampal CA1 region,and immunofluorescence staining was employed to observe the microscopic expression of c-fos-positive neurons in the CA1 region.Results Compared with control group,the target-quadrant dwell time and the platform crossing frequency in Morris water maze test,and discrimination index(DI)and recognition index(RI)in the novel object recognition test,the dendritic spine density of neurons in the hippocampal CA1 region,the c-fos protein expression level and the number of c-fos-positive neurons in the hippocampal CA1 region of mice were all significantly decreased in lead exposure group(P<0.05);and the above mentioned indicators showed no significant difference in EGCG intervention group(P>0.05).Compared with lead exposure group,the learning and memory-related indicators,the dendritic spine density of neurons,c-fos expression and the number of c-fos-positive neurons in mice were all signifi-cantly increased in EGCG+lead exposure group(P<0.05).Conclusion EGCG can improve the learning and memory abilities of lead-exposed mice and increase the number of c-fos-positive neurons in the hippocampal CA1 region.关键词
